The 38-year-old, who won his only major title at the U.S Open in 2012, has appeared in the biennial contest against Europe three times as a player.
He is the first of new U.S captain Keegan Bradley’s assistants to be announced for the event at Bethpage Black, New York, in September 2025.
Bradley said: “Webb is someone who I have long admired and respected as a competitor and friend. He possesses a wealth of team golf experience and knows what it takes to win.”
Simpson, who is a seven-time winner on the PGA Tour and currently stands No. 247 in the official world rankings, said: “I am honoured to be chosen by Keegan to serve as a Vice Captain in 2025.
“Competing in three Ryder Cups will forever be among my career highlights. I have no doubt that Keegan will be a fantastic captain and a tremendous leader, and could not be more excited to get to work as we seek to reclaim the Ryder Cup at Bethpage Black.”
Team Europe are the defending Ryder Cup champions, defeating the Americans in Rome in 2023.
